SR9011 is a synthetic agonist of the nuclear receptors REV-ERB-alpha and REV-ERB-beta, two repressor proteins inside the core circadian clock, and it was made at the Scripps Research Institute in Jupiter, Florida in the laboratories of Thomas Burris and Theodore Kamenecka.

    01Overview

    What SR9011 is

SR9011 was described in the same 2012 Nature paper as its better known sister compound SR9009, which BodyHackGuide covers as SR-9009, and the Scripps authors presented both as tools for shifting the clock and the metabolism it controls and as possible starting points for treating sleep and metabolic disease (PMID: 22460951). No human study of SR9011 is registered: a search of ClinicalTrials.gov on 27 October 2026 for SR9011, SR-9011, SR9009 and stenabolic as interventions returned no trials, so every published result comes from cells, mice, rats or laboratory metabolism work. SR9011 is prohibited in sport at all times, named with SR9009 as an example of a Rev-erb-alpha agonist in section S4.4.1 of the World Anti-Doping Agency 2026 Prohibited List, which went into effect on 1 January 2026.

    How it works

    REV-ERB inside the circadian clock

The mammalian clock runs on a feedback loop in which BMAL1 and CLOCK switch on the Period and Cryptochrome genes, whose proteins then switch BMAL1 and CLOCK back off. REV-ERB-alpha (NR1D1) and REV-ERB-beta (NR1D2) are heme-binding nuclear receptors that act as transcriptional repressors in that loop, and Bmal1 is a direct REV-ERB target gene (PMID: 22460951). An agonist pushes the repressor harder, so clock gene expression and the metabolic genes downstream of it change in timing and amount.

What SR9011 does to the receptor

In a Gal4 chimeric receptor reporter assay in HEK293 cells, SR9011 raised REV-ERB-dependent repression with a half-maximal inhibitory concentration of 790 nanomolar at REV-ERB-alpha and 560 nanomolar at REV-ERB-beta, and it suppressed a luciferase reporter driven by the Bmal1 promoter with full-length REV-ERB-alpha at 620 nanomolar (PMID: 22460951). SR9011 increased recruitment of a nuclear receptor corepressor peptide to the receptor, which is the behaviour expected of a direct agonist, and the Scripps authors reported no activity at other nuclear receptors tested (PMID: 22460951). In cultured suprachiasmatic nucleus explants from a Period reporter mouse, SR9011 suppressed the amplitude of circadian oscillation without changing its period, and the effect reversed when the compound was removed (PMID: 22460951).

Circadian behaviour and metabolism in mice

A single intraperitoneal dose of 100 mg/kg given to mice in constant darkness at circadian time 6, the peak of Rev-erb-alpha expression, removed locomotor activity for the following subjective night, with normal activity returning on the next cycle; the suppression of wheel running was dose-dependent with a half-maximal effective dose of 56 mg/kg, close to the 67 mg/kg for suppression of a REV-ERB target gene in liver (PMID: 22460951). In a 12-hour light and dark cycle the same dose produced only a 1 to 3 hour delay in the start of night-time activity (PMID: 22460951). Over 10 days of twice-daily dosing in Balb/c mice, oxygen consumption rose about 5 percent while movement fell about 15 percent, total food intake did not change, the respiratory exchange ratio did not change, and fat mass fell (PMID: 22460951). Gene expression shifted in the direction of less fat synthesis in liver and white fat and more fuel oxidation in muscle: the lipogenic gene Srebf1 and stearoyl-CoA desaturase 1 fell in liver while the phase of fatty acid synthase expression shifted, the two diglyceride acyltransferase genes for the terminal step of triglyceride synthesis fell in white fat, and carnitine palmitoyltransferase 1b, fatty acid transport protein 1 and two glycolytic enzymes, hexokinase 1 and pyruvate kinase M2, rose in skeletal muscle (PMID: 22460951). All of this is mouse data from injected doses.

Sleep and anxiety-like behaviour in mice

Mice given SR9011 at 100 mg/kg intraperitoneally in the middle of their rest period stayed awake for about 2 hours, with slow-wave sleep broken into more and shorter episodes and REM sleep almost absent for 3 hours, followed by a recovery period after the lights went out; the same dose given during the active period did nothing to sleep patterns (PMID: 25536025). Mice dosed twice daily for 3 to 10 days spent more time in the centre of an open field, more time in the open arms of a plus maze, more time near a novel object and more time in the light compartment of a light and dark box, interacted more with a novel mouse and buried half as many marbles as controls, a pattern read as reduced anxiety-like behaviour with a half-maximal effective dose of 61 mg/kg; REV-ERB-beta-null mice buried more marbles than wild-type mice and did not respond to SR9011, and SR9011 produced no conditioned place preference (PMID: 25536025).

How much of this is actually REV-ERB

The specificity of this compound class is contested. When the Lazar laboratory at the University of Pennsylvania made mice in which both REV-ERB-alpha and REV-ERB-beta could be deleted, the sister compound SR9009 still reduced cell viability, rewired metabolism and altered transcription in hepatocytes and embryonic stem cells that had no REV-ERB protein left, and the authors concluded that the effects of SR9009 cannot be used as a surrogate for REV-ERB activity (PMID: 31127047). That experiment was done with SR9009, and no equivalent double-knockout test of SR9011 was found in the literature retrieved for this guide. Pointing the other way, the anticancer activity of SR9011 in cell lines was abolished when REV-ERB-alpha and REV-ERB-beta were knocked down with short hairpin RNA (PMID: 29320480), and the osteosarcoma study reported that the effects of SR9011 were absent in cells with the receptor knocked out (PMID: 36681687).

    SR9011 side effects and interactions

    Side effects

    Common

    • No human adverse event data exist for SR9011
    • Loss of a full active period in mice given one 100 mg/kg intraperitoneal dose in constant darkness, which the authors attributed to clock disruption rather than toxicity (PMID: 22460951)
    • About 15 percent less movement over 10 days of twice-daily dosing in mice (PMID: 22460951)
    • REM sleep almost absent for 3 hours after one injection in mice, followed by a sleep recovery period (PMID: 25536025)
    • Core temperature about 0.5 degrees Celsius above that of vehicle-treated mice for about 2 hours after one injection, less than the rise typical of wakefulness in the dark (PMID: 25536025)

    Serious events

    • Lower phagocytic activity, mitochondrial respiration, ATP production and metabolic gene expression in primary rat microglia (PMID: 33101272)
    • Deliberate cytotoxicity: SR9011 killed cancer cell lines and oncogene-induced senescent cells by blocking autophagy and de novo lipogenesis, two normal processes in healthy tissue (PMID: 29320480)
    • Part of the activity of this chemical series is off-target, shown for SR9009 in cells lacking both REV-ERB receptors (PMID: 31127047)
    • No human pharmacokinetic, long-term, reproductive or drug interaction data of any kind

    Who should not use it

    Contraindications

Because SR9011 has never been studied in humans, no evidence-based contraindications can be stated (ClinicalTrials.gov, searched 27 October 2026).

Read all the contraindications and cautionsShow less

SR9011 is not an approved medicine: it has no approval in any country, no pharmacopoeial standard and no established human dose.

It is prohibited in sport: section S4.4.1 of the World Anti-Doping Agency 2026 Prohibited List prohibits Rev-erb-alpha agonists at all times and names SR9009 and SR9011 as the examples, and substances in S4.4 are non-specified substances (World Anti-Doping Agency, 2026 Prohibited List).

Sleep and circadian disorders are a concern because SR9011 is designed to change the timing and amplitude of clock gene expression; it suppressed oscillation in suprachiasmatic nucleus explants and removed a full active period in mice in constant darkness (PMID: 22460951), and it suppressed REM sleep for about 3 hours in mice (PMID: 25536025).

SR9009 and SR9011 block autophagy and de novo lipogenesis, which is why they kill cancer cells and senescent cells (PMID: 29320480). Both processes are normal functions in healthy tissue, and no human data describe the consequences of inhibiting them.

SR9011 reduced phagocytic activity, mitochondrial respiration and ATP production in primary microglia, which are immune cells (PMID: 33101272).

Off-target activity is uncharacterized: the sister compound SR9009 acted on cells lacking both REV-ERB receptors (PMID: 31127047), so part of the pharmacology of this chemical series is unexplained.

    08Questions

    Frequently asked questions

    What is SR9011?

SR9011 is a synthetic agonist of REV-ERB-alpha and REV-ERB-beta, nuclear receptors that act as repressors inside the circadian clock, made at the Scripps Research Institute in Jupiter, Florida (PMID: 22460951). In cell reporter assays, SR9011 raised REV-ERB-dependent repression, with half-maximal inhibitory concentrations of 790 nanomolar at REV-ERB-alpha and 560 nanomolar at REV-ERB-beta (PMID: 22460951). SR9011 was built as a research tool for shifting the clock and the metabolism it controls, and it has never been developed as a medicine.

How is SR9011 different from SR9009?

SR9011 and SR9009 were described in the same 2012 Nature paper and differ by one group added at the end of the synthesis, a pentyl urea in SR9011 and an ethyl carbamate in SR9009 (PMID: 22460951). The published SR9011 and SR9009 results overlap heavily: both suppressed REV-ERB target genes in mouse liver, both removed an active period in mice kept in constant darkness, and both raised wakefulness and suppressed REM sleep in mice (PMID: 22460951, PMID: 25536025). The metabolic studies in diet-induced obese mice and the glioblastoma experiments in living animals used SR9009 rather than SR9011 (PMID: 22460951, PMID: 29320480), and so did the one experiment that tested whether the effects survive deletion of both REV-ERB receptors (PMID: 31127047). BodyHackGuide covers SR9011's sister compound as SR-9009.

Has SR9011 been tested in humans?

SR9011 has no registered human study. A ClinicalTrials.gov search on 27 October 2026 for SR9011, SR-9011, SR9009 and stenabolic as interventions returned no trials, and PubMed holds no human pharmacokinetic study, case report or trial of SR9011. All published SR9011 results are from cells, mice, rats or laboratory metabolism experiments, and the mouse results came from injected doses rather than oral ones (PMID: 22460951, PMID: 25536025).

    Is SR9011 banned in sport?

SR9011 is prohibited at all times under section S4.4.1 of the World Anti-Doping Agency 2026 Prohibited List, which prohibits Rev-erb-alpha agonists and gives SR9009 and SR9011 as its examples (World Anti-Doping Agency, 2026 Prohibited List, in effect from 1 January 2026). SR9011 sits in section S4.4, whose substances are non-specified substances, and the World Anti-Doping Agency describes a specified substance as one that potentially allows, under defined conditions, for a greater reduction of a sanction (World Anti-Doping Agency, 2026 Prohibited List). Doping control laboratories have published the phase I metabolites of SR9011 in human liver microsomes and screening methods for both compounds (PMID: 27706103, PMID: 26842585).

What did SR9011 do in animal studies?

One intraperitoneal dose of SR9011 at 100 mg/kg removed the following active period in mice kept in constant darkness, and under a normal light and dark cycle the same dose delayed the start of night-time activity by 1 to 3 hours (PMID: 22460951). Over 10 days of twice-daily dosing, mice given SR9011 used about 5 percent more oxygen, moved about 15 percent less, ate the same amount and lost fat mass, with liver and fat genes for fat synthesis down and muscle genes for fuel oxidation up (PMID: 22460951). In mice implanted for electroencephalography, SR9011 given in the rest period raised wakefulness for about 2 hours and almost abolished REM sleep for 3 hours, and chronic dosing reduced anxiety-like behaviour in six assays (PMID: 25536025). No study has measured strength, endurance or body composition in a human given SR9011.

    Is SR9011 safe?

No human safety data exist for SR9011, and no safety study of it has been registered or published (ClinicalTrials.gov, searched 27 October 2026). In mice given SR9011, the Scripps authors reported no fall in grip strength, continued movement in an open field and normal complete blood counts, and argued that the loss of a full active period in constant darkness was clock disruption rather than toxicity (PMID: 22460951). Laboratory findings on SR9011 that cut the other way include loss of REM sleep for about 3 hours in mice (PMID: 25536025), lower phagocytosis, mitochondrial respiration and ATP production in primary microglia (PMID: 33101272), and the deliberate killing of cancer and senescent cells through blockade of autophagy and fat synthesis (PMID: 29320480). Part of the pharmacology of the SR9011 chemical series is unexplained, because the sister compound SR9009 still acted on cells with both REV-ERB receptors deleted (PMID: 31127047).

    Does SR9011 work through REV-ERB?

SR9011 binds and activates REV-ERB-alpha and REV-ERB-beta in cell assays and increases corepressor recruitment, which is direct agonist behaviour (PMID: 22460951). Several SR9011 effects disappear when the receptors are removed: its anticancer activity was abolished by short hairpin RNA knockdown of both receptors (PMID: 29320480), its effects in osteosarcoma cells were absent in receptor-knockout cells (PMID: 36681687), and its anxiolytic effect was absent in REV-ERB-beta-null mice (PMID: 25536025). The caution for SR9011 comes from its sister compound: SR9009 altered viability, metabolism and transcription in hepatocytes and embryonic stem cells lacking both receptors, and the authors concluded that its effects cannot be read as REV-ERB activity (PMID: 31127047). The same experiment has not been published for SR9011.
