Reported Coluracetam side effects
Human data No human safety data for coluracetam have been published. The one registered human trial, a 6-week randomised double-blind placebo-controlled study of BCI-540 in adults with major depressive disorder and concomitant anxiety, listed safety, tolerability, adverse events, laboratory values, electrocardiograms, vital signs and a physician withdrawal checklist among its secondary outcome measures, but its ClinicalTrials.gov record carries no posted results and no publication of it was found in PubMed (ClinicalTrials.gov NCT00621270). The exclusion criteria of that trial record show what its sponsor treated as a risk. They excluded people with a known hypersensitivity to any cholinesterase inhibitor or cholinergic agonist, people with a history of seizures, people with a history or current evidence of low red cell, haemoglobin, white cell, platelet or reticulocyte counts, and people who would use drugs known to be related to agranulocytosis or aplastic anaemia during the study (ClinicalTrials.gov NCT00621270). Those are study design choices, not findings about coluracetam. Animal observations In the 1996 comparison with tacrine, oral coluracetam at 1 to 10 mg/kg improved learning in AF64A-treated rats "without any significant side effects, like tremor, salivation or hypothermia, which were observed in rats treated with high doses of tacrine" (PMID: 8740080). When injected into the ventricles of rats, coluracetam raised plasma glucose much less than neostigmine, and the authors judged its potency for producing peripheral hyperglycaemia to be pharmacologically negligible (PMID: 10082325). In rats, coluracetam given into the spinal canal or by gavage changed gut function: it raised colonic motility in control animals and relieved stress-induced visceral pain, both through the choline transporter CHT1 (PMID: 30347940, PMID: 30288092, PMID: 36808325). Gut effects of this kind have not been looked for in people. Gaps No published study reports the pharmacokinetics, tolerated dose range, long-term safety, pregnancy safety or drug interactions of coluracetam in humans.
When to continue, reduce, or stop Coluracetam
Mild: continue protocol
Transient mild symptoms (slight GI upset, minor injection-site reaction, brief fatigue) typically resolve within 3-7 days as physiology adjusts. Continue at current dose; monitor weekly.
Moderate: reduce dose
Persistent mild symptoms past two weeks, or new symptoms like sustained mood changes, sleep disruption, or appetite shifts. Reduce dose by 25-50% and reassess at the end of the next 2-week window.
Severe: discontinue + seek care
Any signal of allergic response (rash, swelling, breathing difficulty), unexplained chest discomfort, severe injection-site reaction with infection signs, or vision changes. Discontinue immediately and consult a licensed practitioner.
Coluracetam contraindications
No human safety record. Coluracetam has no published human safety data of any kind, and the single registered trial posted no results (ClinicalTrials.gov NCT00621270). Every statement about its effects below rests on rodent or cell experiments. Cholinergic drugs. Coluracetam raises acetylcholine synthesis and release in rodent tissue by acting on the choline transporter (PMID: 18446264, PMID: 18461273). Drugs that add to cholinergic tone, such as cholinesterase inhibitors and cholinergic agonists, act on the same system from a different direction, and the registered trial excluded people with a known hypersensitivity to either class (ClinicalTrials.gov NCT00621270). Seizure history and blood count abnormalities. The registered trial excluded people with a history of seizures and people with low or abnormal blood counts (ClinicalTrials.gov NCT00621270). No published coluracetam data explain those exclusions. Not an approved medicine. Coluracetam is not approved in the United States or the European Union, and FDA has stated that it has not approved racetam drugs, naming coluracetam among them (Department of Justice press release posted by FDA, 30 October 2023).
Why vendor quality drives Coluracetam side-effect frequency
Research-compound communities consistently report that adverse-event frequency tracks with source quality. Three reasons: (1) impurity profiles vary by lab (residual synthesis byproducts can trigger reactions); (2) mislabeled mass means actual dose can be 50-150% of stated dose; (3) reconstitution-grade water and capping methodology affect long-term storage stability. COA-verified vendors with published HPLC data minimize all three failure modes. Check a vendor's COA record and Trust Score before you buy.

Coluracetam side effects FAQ
What are the most common Coluracetam side effects?
Human data No human safety data for coluracetam have been published. The one registered human trial, a 6-week randomised double-blind placebo-controlled study of BCI-540 in adults with major depressive disorder and concomitant anxiety, listed safety, tolerability, adverse events, laboratory values, electrocardiograms, vital signs and a physician withdrawal checklist among its secondary outcome mea
Are there severe side effects to watch for with Coluracetam?
Severe reactions are rare in published research but can occur. Watch for: allergic-type symptoms (rash, swelling, breathing difficulty), unexplained chest discomfort, infection signs at injection sites, vision changes, or severe persistent mood/sleep disruption. Any of these warrants immediate discontinuation and consultation with a licensed practitioner. The Severity Tiers section above triages mild → moderate → severe response patterns.
Does Coluracetam have contraindications?
No human safety record. Coluracetam has no published human safety data of any kind, and the single registered trial posted no results (ClinicalTrials.gov NCT00621270). Every statement about its effects below rests on rodent or cell experiments. Cholinergic drugs. Coluracetam raises acetylcholine synthesis and release in rodent tissue by acting on the choline transporter (PMID: 18446264, PMID: 1846
How can I minimize Coluracetam side effects?
Three biggest levers: (1) Start at the Beginner-tier dose for 2-3 weeks before escalating, which gives time to detect adverse signals at minimum effective dose. (2) Source from COA-verified vendors only: impurity profiles and mislabeled compounds drive a disproportionate share of reported side effects in research-compound communities. (3) Maintain adequate cycle off-time when applicable: receptor-saturating compounds benefit from 4-6 week breaks. The vendor section below lists the cheapest in-stock Coluracetam sources we track, each linked to its Trust Score (COA verification is 30% of the score).
