    Side Effects & Safety

    In the 12-week phase 1 multiple-ascending-dose study in 100 participants, the most common treatment-emergent adverse events were decreased appetite in 19 percent, headache in 12 percent, fatigue in 11 percent and COVID-19 in 11 percent; gastrointestinal events were infrequent, with diarrhea in 10 percent, nausea in 8 percent and vomiting in 4 percent. Most events were mild, there were no deaths, and the single serious adverse event was judged unrelated to the drug (PMID: 41559929). In the 48-week phase 2 trial, nausea and fatigue were the most common adverse events and both rose with dose, reaching 64 percent for nausea and 46 percent for fatigue in the highest-incidence groups against 14 percent and 12 percent on placebo (PMID: 41207310). A network meta-analysis noted that gastrointestinal adverse events, nausea, vomiting and constipation, were more common with high doses of amylin-based therapies generally (PMID: 42175595).

    Contraindications
