# PDRN

Skin & Hair: https://www.bodyhackguide.co/wiki#cat-skin-&-hair

Status

Phase 4,, not approved for human use

Evidence

Not graded on a research bench yet

PDRN (Polydeoxyribonucleotide) is a biotechnological compound derived from salmon sperm DNA (Oncorhynchus mykiss) through extraction, purification, and fractionation.

Reviewed 28 October 2026

01 Overview

## What PDRN is

It consists of polynucleotide chains with molecular weights ranging from 50 to 1500 kDa. PDRN is widely used in Asian aesthetic medicine (particularly South Korea) for skin rejuvenation, wound healing, and hair loss treatment. It is available as an injectable sterile solution (Placentex Integro) licensed in several countries, as well as topical formulations and cosmetic creams. PDRN's mechanism centers on adenosine A2A receptor activation, stimulating collagen synthesis, cellular proliferation, and anti-inflammatory pathways. It is one of the few compounds in this catalog with a substantial published clinical (Phase 4) evidence base in aesthetic applications.

### How it works

PDRN exerts effects through two primary mechanisms: (1) Adenosine A2A receptor agonism — PDRN is hydrolyzed to deoxyadenosine fragments that activate A2A receptors on fibroblasts, endothelial cells, and macrophages. A2A activation stimulates VEGF production (angiogenesis), collagen type I and III synthesis, and anti-inflammatory cytokine profiles (increased IL-10, decreased TNF-α). (2) Salvage pathway activation — DNA-derived nucleotides from PDRN are incorporated into the tissue nucleotide salvage pathway, providing precursors for DNA repair and cellular proliferation in wound healing. Together, these mechanisms accelerate fibroblast proliferation, neocollagenesis, angiogenesis, and tissue regeneration.

### Benefits listed

- Wound healing acceleration - enhanced angiogenesis, fibroblast proliferation, and collagen synthesis
- Diabetic foot ulcer healing - supported by a randomized, double-blind, placebo-controlled trial (complete healing 37.3% vs 18.9% for placebo) [PMID:24483158]
- Skin rejuvenation - clinical studies suggest improved texture and elasticity, though a 2024 systematic review found the aesthetic evidence base inconsistent [PMID:39198280]
- Hair loss - preliminary evidence only; a small uncontrolled study in female pattern hair loss reported improved hair count and thickness (not established for androgenetic alopecia) [PMID:25524027]

06 Chemistry

## Chemistry and structure

Molecular mass

No single value - PDRN is a heterogeneous mixture of deoxyribonucleotide polymers (chain lengths ~50-2000 bp, roughly 50-1500 kDa) rather than one defined molecule [PMID:19860658].

Half-life

Short (oligonucleotides degraded rapidly; tissue effects persist longer)
