Unlike testosterone or hCG, kisspeptin acts at the very top of the axis and recruits the body's own GnRH pulse generator. It has been studied in human clinical trials for IVF egg-maturation triggering, hypogonadism, hypothalamic amenorrhea and sexual desire, but it is not an approved drug and is sold strictly for research use only (RUO).

### What to Expect

What is KSPTN?

It is a potent agonist of the kisspeptin receptor KISS1R (GPR54).

What does kisspeptin actually do?

It acts upstream of the reproductive axis. Kisspeptin stimulates GnRH neurons in the hypothalamus, which drives the pituitary to release LH and FSH, which signal the gonads to make testosterone (men) or estrogen and progesterone and to trigger ovulation (women). According to PubMed, an IV dose of kisspeptin-10 potently raises LH and, in men, testosterone in healthy volunteers \[PMID:17323132\].

Is kisspeptin FDA-approved?

No. Kisspeptin is investigational. It has been studied in numerous human clinical trials - for IVF egg-maturation triggering, hypogonadism, hypothalamic amenorrhea and sexual desire - but it is not an approved drug, and material sold to consumers is labeled research use only.

Can I inject it subcutaneously like other peptides?

There is no validated subcutaneous self-dosing protocol. Every human study of kisspeptin-10 used intravenous administration because the peptide is cleared within minutes. Kisspeptin-54 (a longer form) lasts longer and has been given subcutaneously in IVF trials. Community subcutaneous use of kisspeptin-10 is anecdotal and not supported by pharmacokinetic data.

Will more kisspeptin give a bigger hormonal response?

Not necessarily. The kisspeptin receptor desensitizes with continuous or high-dose exposure, which can blunt the LH response. The natural signal is pulsatile, so overdosing tends to reduce, not amplify, the effect.

How is kisspeptin different from gonadorelin or hCG?

Gonadorelin is GnRH itself, one step further down the axis, and hCG mimics LH directly at the gonad. Kisspeptin acts one step higher, on the GnRH neurons, so it recruits the body's own GnRH pulse generator. In IVF it has been studied as a gentler oocyte-maturation trigger than hCG, with lower ovarian-hyperstimulation risk \[PMID:28393578\].

What is the difference between kisspeptin-10 and kisspeptin-54?

The KISS1 gene makes a 54-amino-acid peptide (kisspeptin-54, or metastin) that can be cleaved into shorter fragments (kisspeptin-14, -13 and -10) sharing similar receptor potency \[PMID:18193176\]. Kisspeptin-10 is the short decapeptide most commonly sold as research material; kisspeptin-54 is longer-acting and used in several clinical trials.
